چکیده

OBJECTIVE:We evaluated the effectiveness of spermwashing using the discontinuous two-layer density gradient centrifugation method to remove antisperm antibodies attached to the sperm surface. METHOD: We prospectively enrolled sixty-six men with unexplained infertility who were seeking evaluation. Each patient delivered one semen specimen for the study. We determined antisperm antibody levels using the direct immunobeads test. Specimens were classified into two groups according to the pre-washing levels of antibody-bound spermatozoa: group 1 (low antisperm antibodies levels, immunobeads test,20%; n 1⁄4 54), and group 2 (high antisperm antibodies levels, immunobeads test $20%; n 1⁄4 12). Sperm washing was carried out using the discontinuous two-layer colloidal density gradient centrifugation method. Preand post-wash levels of antisperm antibodies were compared in the groups. RESULTS: The preand post-wash percentage of spermatozoa with antisperm antibodies attached to their surface was 11% and 6.5% in group 1 (mean difference 1⁄4 40%; p , 0.01), and 30% and 19.5% in group 2 (mean difference 1⁄4 36.8%, p 1⁄4 0.02), respectively. The effectiveness of density gradient centrifugation in removing antisperm antibodies was not different between the groups, but individual variations from 252.3% to 23.9% were observed. CONCLUSIONS: Sperm washing by density gradient centrifugation is an overall effective method to remove antibodies bound to sperm membranes, regardless of the levels of antisperm antibodies in the neat semen. Due to an inter-individual variation in the effectiveness of the method, we recommend that each patient be tested before applying sperm washing by density gradient centrifugation in intrauterine insemination.

Comparison of two sperm processing techniques for low complexity assisted fertilization: sperm washing followed by swim-up and discontinuous density gradient centrifugation

OBJECTIVE This study aimed to assess and compare sperm motility, concentration, and morphology recovery rates, before and after processing through sperm washing followed by swim-up or discontinuous density gradient centrifugation in normospermic individuals. METHODS Fifty-eight semen samples were used in double intrauterine insemination procedures; 17 samples (group 1) were prepared with sper...

Stallion Sperm Selection by Density Gradient CentrifugationInvolving a Double Layer Colloid: Effects on Sperm Subpopulation Dynamics in Fresh and Stored Semen

Sperm selection techniques have became an important tool to improve sub fertile ejaculates. The main aim of this study was to assess the dynamics of motile sperm subpopulations of ejaculates from subfertile stallions subjected to density gradient centrifugation (DGC) involving a double layer colloid [DGC2], before and after storage for 24 and 48 hours. Ejaculates from eight stallions with ferti...

Comparative study of four different sperm washing methods using apoptotic markers in ram spermatozoa.

The accurate measurement of semen fertilizing potential is of great importance in determining the acceptability of processed semen for breeding purposes. A good sperm preparation technique results in a sample with high viability and motility and also takes into account other parameters such as the capacitation and apoptotic state which could compromise the ability to fertilize an oocyte. In thi...

P-12: Effect of Centrifugation on Levels of Stress Oxidative of Human Spermatozoa

Background: Cryopreservation has adverse effect on human spermatozoa via exposure to physical and chemical stress that may lead to alter in reactive oxygen species (ROS) levels. During sperm preparation or cryopreservation, centrifugation as a physical stress may effect on sperm quality probably with stress oxidative production and membrane damage. Therefore the aim of this study was to evaluat...

Apoptosis and necrosis in human ejaculated spermatozoa.

BACKGROUND Features of both apoptosis and necrosis have been reported in ejaculated human spermatozoa. This study examines the relative contribution of these two modes of cell death to the demise of these terminally differentiated cells. METHODS Sperm fractions were prepared from aliquots of semen samples from young normozoospermic donors by simple washing from seminal plasma, by discontinuou...
